The multiplex tissue image of this month showcases the spatial expression pattern of acinar cell-regulating basic helix-loop-helix family member a15 (gene: BHLHA15) in exocrine glandular structures of the salivary gland.

Salivary gland acinar cells are glandular cells that produce saliva. Enzyme-rich secretions from serous acini and mucin-rich secretions from mucus acini combine to form the final saliva. Through the use of multiplex immunohistochemistry (mIHC/IF) and the salivary gland panel, BHLHA15 - a transcription factor involved in the regulation of acinar cell function - is specifically localized (green) to the nucleus of serous acini (cyan) within the salivary gland. Other visualized structures include large ducts (red) and small ducts (magenta).

The spatial expression data from salivary gland is further supported by the single-cell RNA sequencing data for BHLHA15 available in the Single Cell resource at the Human Protein Atlas showing enhanced expression in salivary acinar cells.
